Fowler is best known for his time with the Chicago Cubs, with whom he won the World Series Championship in 2016. He also played for the Houston Astros, Colorado Rockies, St. Louis Cardinals, and Los Angeles Angels.

The Colorado Rockies drafted Fowler in the MLB Draft of 2004 in the 14th round. He made his Major League debut with the Rockies on September 2, 2008. Fowler played for the Rockies until December 2013, when he was traded to the Houston Astros for pitcher Jordan Lyles and outfielder Brandon Barnes. During the 2014 season, Dexter Fowler hit .276 in 116 games for the Houston Astros.

In January 2015, Dexter Fowler was traded to the Chicago Cubs for pitcher Dan Straily and infielder Luis Valbuena. The Cubs won 4-0 over the Pittsburgh Pirates in the 2015 National League wild-card game, and for the first time in seven years, the Chicago Cubs entered the postseason after finishing the season with a 97-65 record. In 2016, Dexter Fowler was selected to represent the Cubs in his first All-Star Game along with six other teammates.

On October 25, 2016, Fowler and other teammates, including Carl Edwards Jr., Addison Russel, and Jason Heyward, became the first African-Americans to play for the Chicago Cubs in a World Series Championship game. In the same year, Dexter Fowler opened Game 7 of the World Series by hitting a home run. This made him the first baseball player to open a World Series Game 7 with a home run.
