The 1992 NPC Southern States Championship was Dexter Jackson's first NPC (National Physique Committee) contest, and he finished third in it. After the competition, he took three years off and devoted himself entirely to training and diet. He wanted to bring the best version of himself to win the Pro card.

Dexter Jackson took to the stage again after a three-year break. In 1995, he made a comeback and hosted the NPC USA Championship's Light-Heavyweight division. When he won the Light-Heavyweight and general divisions of the North American Championships in 1998, everyone was taken aback. The Pro card was eventually given to him.

At the 1999 Night of Champions, Arnold Classic, and Mr. Olympia contests, Dexter Jackson made his professional debut and finished third, seventh, and ninth, respectively. In 2003, he came third in Mr. Olympia and won the 2003 Show of Strength Pro Championship.

Dexter Jackson finished third at the 2007 Mr. Olympia, and many detractors predicted that he wouldn't finish any higher. He defeated reigning two-time Mr. Olympia Jay Cutler on September 27, 2008, to become the title's 12th winner and just the second person to win both the Arnold Classic and Mr. Olympia titles in the same year. Along with Samir Bannout (1983), Shawn Rhoden (2018), Brandon Curry (2019), and Chris Dickerson (1982), Dexter Jackson is the only person to have won the Mr. Olympia title once.

Dexter Jackson had a fantastic year in 2008, taking home victories at Mr. Olympia, Arnold Classic, Russian Grand Prix, New Zealand Grand Prix, and Australian Pro Grand Prix VIII. He placed third in the 2009 Mr. Olympia contest. He finished fourth in the Mr. Olympia competition in 2012, then amazed everyone by taking first place in IFBB Masters Olympia at the age of 43.
