Dia Mirza was born in the Indian city of Hyderabad, Telangana, as Dia Handrich. Mirza was enrolled at Nasr School, a girls' day school in Khairtabad, after attending Vidyaranya High School, a co-ed school.

Her parents split when she was four and a half years old. Her mother took her stepfather's surname after marrying Ahmed Mirza, a Muslim man from Hyderabad. In 2003, he passed away.

In 2000, she finished as second runner-up in the Miss India pageant before winning the Miss Asia Pacific title.
'Rehnaa Hai Terre Dil Mein' was her first film. It flopped at the box office but gained a cult following among Indian youth. 'Alag-He is different', 'Dum', 'Deewanapan', 'Tumko Na Bhool Paayenge', 'Tumsa Nahin Dekha – A Love Story', 'Parineeta', 'Dus', 'Lage Raho Munnabhai', and 'Salam Mumbai' were among her subsequent films.
