Joseph William Calzaghe was born in London, England. Joe was born to a Sardinian father, Enzo Calzaghe, and a Welsh mother, Jackie. Joe has two sisters, Melissa and Sonia. Joe's family lived briefly in Sardinia before moving back to London and then to Newbridge, South Wales.

Calzaghe made his professional debut in 1993 against Paul Hanlon. He went on to win all his subsequent matches. Calzaghe earned the British championship in the super-middleweight category in 1995 after beating the previous champion, Stephen Wilson. In 1996, Joe defended his British title from the undefeated Mark Delaney. Joe's victories in 1997 included opponents Carlos Christie, Tyler Hughes, and Luciano Torres.
Joe regards his bout with British boxer Chris Eubank on October 11, 1997, for the WBO super-middleweight title as the toughest fight of his life. He won his bout against Branko Sobot with a knockout in the third round in 1998.
Calzaghe was the first person to defeat Juan Carlos Gimenez Ferreyra with a total knockout after nine rounds. He defended and retained his WBO title against multiple opponents, including Robin Reid (1999), British boxer David Starie (2000), Omar Sheika (2000), and erstwhile WBC world champion Richie Woodhall (2000) across his career. Joe also beat the hitherto unbeaten Mario Veit in 2001 and against Tocker Pudwill with a total knockout in 2002.
Former champion Byron Mitchell dealt Joe his first career knockdown in 2003. Joe recovered to beat Mitchell with a total knockout in the second round to retain his title.
Joe won the IBF super-middleweight championship title in 2005 from Jeff Lacy. At the same time, he also won the first Ring super-middleweight title. In 2007, Calzhage won against Kessler in a unification match for the WBC and WBA super-middleweight titles while retaining his WBO and Ring titles.
Joe Calzaghe went up against Peter Manfred Jr. in 2007 and handed over the IBF super-middleweight championship.
