The New York Giants drafted Amani Toomer as the 34th overall pick in the second round of the 1996 NFL Draft. He played as a punt and kickoff returner in his first three years with the New York Giants. Toomer set a franchise record in his debut NFL game against the Buffalo Bills with an 87-yard punt return for a touchdown, which was the opening game of the 1996 season. In the 1997 season, he appeared in all 16 games for the team and returned 47 punts for 455 yards with an average of 9.7 yards per return. The following season, Toomer played all 16 games for the Giants and returned 35 punts for 252 yards, averaging 7.1 yards per return. He scored five touchdowns and registered 27 catches for 360 yards. In an impressive comeback by the Giants, Toomer caught a 37-yard touchdown pass from Kent Graham with 48 seconds left to win against the undefeated Denver Broncos.
In the 1999 season, Toomer was a starting wide receiver in all 16 games for the Giants and broke the team record for receptions in a season with 79. His best season was 2002, where he had eight receiving touchdowns with 82 receptions and 1,343 receiving yards. In the 2007 season, Toomer played all 16 regular season games scoring three touchdowns and totaling 59 receptions for 760 yards. The New York Giants went on to win Super Bowl XLII, defeating the New England Patriots. The 2008 season was the last NFL season for Amani Toomer, and he played all 16 regular season games, starting in 12 of them. The wide receiver caught 48 passes for 580 yards and scored four touchdowns. In 2009, Toomer signed a one-year contract with the Kansas City Chiefs but was later released.
In 2021, Amani Toomer participated in Bike HSS, a fundraiser to help support the research and needs of the Hospital For Special Surgery in New York. He also started the Amani Toomer Foundation.
