His parents enrolled him in Eastern Commerce Collegiate Institute for his high school education. He was a vital member of the school basketball team, leading it to the Ontario provincial championships in 1995-1996.

After graduating from high school, Jamaal Magloire attended the University of Kentucky in 1996. During his college career in basketball, he was chosen to the SEC All-Freshman team. As a sophomore, Magloire won the NCAA championship in 1998. With 268 blocked shots, he concluded his college career as Kentucky's All-Time Leader. After graduating from the University of Kentucky, Jamaal Magloire got selected by the Charlotte Hornets in the 2000 NBA draft. For the first two seasons, he was kept in reserve. In 2003–2004, he was chosen to the Eastern Conference All-Star Team, the NBA's second Canadian All-Star, following Steve Nash. Magloire had a strong game, leading the Eastern All-Stars with 19 points and eight rebounds in 21 minutes.

In 2005, Jamaal Magloire transferred to the Milwaukee Bucks, where he stayed for a year. Next, he joined the Portland Trail Blazers in 2006-2007, after which he decided to become a free agent. As a free agent, he signed a contract with the New Jersey Nets in 2007 and was waived off by the team in 2008. After that, Magloire joined the Dallas Mavericks and Miami Heat.

With Miami Heat, Jamaal Magloire proved to be a valuable player in the center position due to his experience and knowledge. He was re-signed by the team in the 2009-2010 and 2010-2011 seasons. Miami Heat even reached the 2011 NBA finals with him on the team. However, they lost the match to Dallas Mavericks.
