Katharine Meyer was born in June 1917 in New York City to Agnes Elizabeth and Eugene Meyer, who came from an upper-class family. Her father was a banker who became the chairman of the Federal Reserve. In 1933, her father purchased The Washington Post at a bankruptcy auction. Her mother was a Republican Party activist, an art lover, a bohemian intellectual, and a political activist.

Her father was an Alsatian Jew, while her mother was a Lutheran. Florence, Eugene III (Bill), Ruth, and Elizabeth Meyer were her siblings. Florence Meyer, her older sister, was a prominent photographer and the wife of Oscar Homolka, an actor. Florence Meyer Blumenthal, her father's sister, founded the Prix Blumenthal.

Meyer started as a reporter with the Post in 1938. In 1946, Phil Graham (her husband) was named publisher of the Washington Post. In September 1963, however, she was president and de facto publisher of the paper after the death of Philip Graham. From 1969-1979, she was the publisher, and from 1973 -1991, she was the board's chairwoman. In 1972, as CEO of the Washington Post, she became the first female Fortune 500 CEO. Graham emphasized her own lack of faith and doubt in her understanding in her memoir.

Graham employed Benjamin Bradlee as an editor and asked Warren Buffett for financial guidance; Buffett eventually became a substantial shareholder and the company's eminence grise. From 1979 until 2000, her son Donald served as a publisher.

Graham led the Post during a critical juncture in its history. The Washington Post was instrumental in exposing the Watergate story scandal, which led to president Richard Nixon's resignation.
