To investigate the container's disappearance, the physicist Vittoria and Landon's daughter travels to Vatican city. The initial investigations of Landon disclose the four cardinal murders that took place mimicking the earth elements: earth, water, fire, and wind.

He finds the victims being branded with ambigrams. Victoria took the hostage, and Langdon continued to pursue his search in finding the canister and, at the same time, determined to pursue his colleague. Later, Landon found her in St. Peter's basilica catholic church.

The book's first edition had many inaccuracies regarding the places in Rome and incorrect usage of the Italian language. A few of the language problems were addressed in the later editions. Apart from the book's explicit intro, it depicts several fictional experts describing the matters in science, history, and technology in which inaccuracies were pointed out.

One of the examples is regarding the antimatter discussion; the book states that antimatter can be made in practical and valuable quantities and is it has limitless power.
